Check engine light Toyota Rav 4

Check engine light on in my Toyota RAV 4, 6 cylinder. Tune up did not fix. Shows misfire in cylinder #1. Could this also be the loose hose or the gas cap? Mechanic cannot figure out the issue.

Thanks for your response. I do know he checked the cylinder. Previous scans of the system (lights going on/off for about a year along with 4WD & anti-skid) showed other codes I cannot name.
