Rea differential Upgrade

I want to swap my 1989 chevy step side rear
differential with 5 lug drum brakes for a 2000
chevy sport side with 6 lug disc brakes. Will it
work?

I think it is possible. Take measurements and compare the spring seats. You may have to have a custom drive line built. You would want to find a 6 lug conversion for the front so that you can use matching wheels front and back.
