Hi - I have 2014 Mercedes-Benz E350 4dr Sdn E 350 Sport 4MATIC AWD with a flat tire. Would i have to replace one tire or all 4 tires?

Depends on how worn the other three are. If relatively new. an exact match new should be OK. If more than 50% worn, replace all 4. If you really want to gamble, find a used tire nearly exact with similar treadwear and go with it. All hinges on what your plans are for the car's future.... and yours.
