Can I fit 35x12.5 on 18" wheels on my 1998 single cab dodge 2500?

Those tires would be 1.3" wider and 2.2" taller. I can't say if they would fit so I would recommend having the tire shop try one on to check the clearance.

If you were to guess, do you think they would with a 2" coil spring spacer in the front and left the rear the same as it is? This is my truck now with the 16s

I would try measuring things to see. I do think it is likely that you will have some clearance issues in the front.
