When the engine is running you have additional force applied to the master cylinder via the brake booster. Typically a problem with the MC can result in ALL of the brakes locking up. If it's only one brake locking up, the problem may be in that line or caliper. However, it may be a combo of the two, a MC or Booster that's failing and causing an increase in hydraulic pressure to all of the lines, but you are feeling it first in one line in particular due to a failing brake hose or caliper. Your brakes need a thorough inspection.
